Niger Tornadoes set the tone for the season with a 2-1 away victory over Kwara United at the Ilorin Township Stadium. Joseph Godstime's 32nd-minute strike made him the first player to score away from home in the 2024/2025 NPFL season.

In Umuahia, Remo Stars spoiled Imama Amapakabo's return to club management with a 2-0 win over Abia Warriors. The Oriental Derby in Owerri lived up to expectations as Enyimba FC came from behind to beat Heartland FC 3-1 at the Dan Anyiam Stadium.

Newcomers Ikorodu City FC were given a baptism of fire, losing 3-0 to Kano Pillars, while Plateau United edged Katsina United 1-0 in Jos.
